Encyclopedia > DNA primase

DNA primase is form of RNA polymerase and a product of the dnaG gene. Primase binds to the DNA helicase forming a complex called the primosome. Primase is activated by DNA helicase where it then synthesizes a short RNA primer approximately 11 ±1 nucleotides long, to which new nucleotides can be added by DNA polymerase. Primase is of key importance in DNA replication because no known DNA polymerases can initiate the synthesis of a DNA strand.

A primer is a short RNA segment that is complementary to a DNA segment, and is necessary to begin DNA replication.

BACTERIAL PRIMASE HOME PAGE (1698 words)
Primase is required because DNA polymerases cannot initiate polymer synthesis; DNA polymerases can only elongate polymers that have already been initiated.
The ts mutants that arrest DNA synthesis immediately after raising the temperature are called quick-stop mutants and are thought to indicate that the gene plays a role in the elongation phase of DNA replication.
The bacterial primases motif is the hinge polypeptide between subdomains 1 and 2 and there is a Hydrogen bond between Arg146 from motif 2 in sub1 and Asp347 from motif 6 in sub2 may stabilize the subdomain 1-2 interface.
