Desmoplakin staining is almost entirely restricted to the apicolateral junctional complex (C), and staining of the cytokeratin network is low throughout the cell, except for moderate staining near the apical and basal cell surfaces (F).
Desmoplakin immunostaining (green) is concentrated in the apicolateral region, but additional staining extends further down the lateral cell surfaces in regions adjacent to the nucleus (red).

The carboxyl terminal domain of desmoplakin associates with IF networks while the desmoplakin amino-terminus is required for association with desmosomes.
To determine if the desmoplakin amino terminal domain could associate with the desmosomal cadherin-plakoglobin complex, L-cell fibroblasts were used to establish stable cell lines that co-expressed desmoglein (Dsg1), plakoglobin and an amino-terminal desmoplakin polypeptide (DP-NTP).
Deletion analysis of plakoglobin suggested that the central armadillo repeats of plakoglobin were sufficient for the association of the plakoglobin-cadherin complex with desmoplakin, since deletion mutants of plakoglobin lacking either the amino or carboxyl terminus both co-immunoprecipitated with DP-NTP.
