Agito is the first Kamen Rider series to feature the three main types of Riders: the mystical Rider (one that draws power from ancient technology or mystical methods), the organic Rider (one whose anatomy changes to become a Rider), and the high-technology Rider (one who is either a cyborg or uses advanced technology to fight, of which majority of the Classic Kamen Riders). Plot summary A man named Shoichi Tsugami seemingly has no memory. He doesn't know who he is, where he came from, or how he came upon his peculiar circumstances. Tsugami, seemingly for no reason, transforms into a powerful superhuman, Agito, whenever in the presence of the beings referred by the police as the "Unknown", a race of powerful monsters that have been causing murders around Tokyo, targeting certain people as their prey. In response, the police department unleash their newest weapon: the G3 powersuit, originally developed to fight against the "Unidentified Lifeforms", the enemies of Kamen Rider Kuuga. G3 and Agito don't know whether they should join up and defeat the Unknowns, their common enemy, or to combat each other, keeping the mysteries that entwine them separate. Ultimately, even more mysteries unfold, with the appearance of Kamen Rider Gills, who is on a search to uncover why his father committed suicide. These mysteries and others collide, as the true nature of Agito would ultimately determine the fate of humanity.

Shoichi uses the Alter Ring to transform into Kamen Rider AgitΩ, an ancient and powerful warrior whose abilities are constantly evolving. In the transformed state, Shoichi possesses the Wiseman's Monolith - a relic of unknown origin and purpose - embedded into his armor. When his crest horns open, AgitΩ's maximum power is unleashed. When in need of greater abilities, the Alter Ring accesses additional powers. His real name is Tetsuya Sawaki.
- Makoto Hikawa / Kamen Rider G3 (氷川 誠 / 仮面ライダーG3, Hikawa Makoto / Kamen Raidā Jī Surī?): A cop assigned to the G3 Unit of Tokyo's metropolitan police to investigate the rash of eerie supernatural murders. He was assigned as the operator for the G3 System, originally developed to combat the Unidentified Lifeforms (from Kamen Rider Kuuga), but now recommissioned to combat the Unknown. Aside from his apparent klutziness and naiveté, Hikawa's frank yet earnest personality is his defining quality. Nevertheless, he is a man full of integrity, but because he is a regular human being, the "Unknowns" usually ignore him.
- Ryo Ashihara / Kamen Rider Gills (葦原 涼 / 仮面ライダーギルス, Ashihara Ryō / Kamen Raidā Girusu?): Formerly a star swimmer of Jyohoku University, Ashihara’s future as a swimmer almost came to an end after a near-fatal car accident. After he healed, he started swimming again, but he has undergone a metamorphosis, enabling him to transform into Kamen Rider Gills. However, the change resulted in the immediate alienation of all those closest to him, and its this loneliness fueled his lust for battle, which in turn makes Gills increasingly feral. His ferocity is so great, that Gills comes across as a brutal savage in the eyes of those whom witness him in battle; brandishing wildly his claws and tentacles at his adversaries. Nevertheless, his transformation is increasingly unstable, causing accelerated cellular decay.
- Yukina Sawaki (沢木 雪菜, Sawaki Yukina?): A young woman psychic, who committed suicide after discovering she was becoming an AgitΩ. She was involved in paranormal research conducted by Professor Kazaya at Jyohoku University. She left behind a younger brother who would become the Masked Rider AgitΩ.
- Mana Kazaya (風谷 真魚, Kazaya Mana?): The niece of Yoshihiko Misugi who Shoichi lives with, possessing telepathic powers for mysterious reasons. She was named after mana by her father.

- Sumiko Ozawa (小沢 澄子, Ozawa Sumiko?): The genius behind the G3 System. Her position within the G3 unit is the systems monitor engineer, which monitors the condition of the operator and the G3 suit during combat from the G-Trailer mobile command base. She was in constant rivalry with Tōru Hōjō.
- Takahiro Omuro / Kamen Rider G3 Mild (尾室 隆弘 / 仮面ライダーG3マイルド, Omuro Takahiro / Kamen Raidā Jī Surī Mairudo?): He helps Ozawa at the G-Trailer mobile command base.
- Tōru Hōjō (北條 透, Hōjō Tōru?): A fellow cop and Hikawa's rival, Hōjō's brilliant and talented in his profession as well as crafty and underhanded, such as his first attempt to capture AgitΩ. He is also the creator of the Victory One System (V1), which was meant to rival the G3 System. Despite his attitude, he has respect for Hikawa; as well as admiration for Ozawa.
- Tetsuya Sawaki (沢木 哲也, Sawaki Tetsuya?): The real Shoichi Tsugami is a man resurrected by the Overlord due to his passive involvement in the death of the first AgitΩ, Yukina Sawaki. In reality, he was romantically involved with Yukina and tried to stop her from committing suicide, but he ultimately allowed her to plummet to her death. He then take the name of Tetsuya Sawaki as he stated to the Overlord that he has give up the name of "Shouichi Tsugami".
- Sakiko Mikumo (三雲 咲子, Mikumo Sakiko?, 1-6): In charge of the O-Parts Research Department, her group research the items from old civilizations long lost. By chance, she and her group found the puzzle that held the Overlord's DNA and unlocked it. With no idea of its true nature, she had the 30,000 old DNA synthesized, resulting with the Overlord of Darkness being reborn as well as the division being disbanded upon his escape.
- Dr. Azuma Kunieda (国枝 東, Kunieda Azuma?, TVSP): A doctor who nursed Shoichi back to health after he washed ashore. Sometime after, Dr. Kuneida would place him in the care of Professor Yoshihiko Misugi.
- Shiro Mizuki / Kamen Rider G4 (水城 史朗 / 仮面ライダーG4, Mizuki Shirō / Kamen Raidā Jī Fō?, Movie): One of the military officers guarding the GA psychic educational facility when the Formica Pedes Lords attacked. Though he would survive the ordeal, he would be left a jaded man, his pride scarred. He began living as one who would embrace death as the true means of becoming stronger. He would eventually volunteer as operator for the G4 suit and eventually clash with G3-X operator Hikawa, to put their ideals to the test on the battlefield.
- Risa Fukami (深海 理沙, Fukami Risa?, Movie): From the GA, assigned to the G3 Unit. She copy the data from Ozawa's computer relating to the G4. Once she got what she needed, she quits the G3 Unit and creates the G4 suit, intend on perfecting it so it can be mass-produced.
- Sayaka Kahara (加原 紗綾香, Kahara Sayaka?, Movie): A young lady with the psychic ability of clairvoyance. She came from a wealthy family and lived relatively well until she was orphaned during a car crash, which killed all but Sayaka herself. She inducted into the research study at the GA psychic educational facility as a candidate psychic for the G4 project.
- Rei Motoki (本木 レイ, Motoki Rei?, Movie): A young boy gifted with psychic ability. He, like most children in the GA Psychic Facility, was orphaned at a young age.
- Kana Okamura (岡村 可奈, Okamura Kana?, 47-50): A part-time chef, but was unable to cook proper and was fired until Shoichi helped her out. Her father was a chef, a victim of a Unknown attack.
- Risa Mizuhara (水原 リサ, Mizuhara Risa?, 47-50): She was a young runner for her high school team, but was hurt and ostracized by her former friends. Taking the identity of the Scorpion, Risa raced recklessly to forget her past.

The Lords Lord (ロード, Rōdo?): The major antagonists of Agito, referred by the police department as Unknown (アンノウン, An'nōn?), to differ them from the Unidentified Lifeforms (Grongi Tribe) that attacked humanity two years ago. The Lords are a group of powerful disciples serving under the OverLord, who created them from his body. All of their tribes are separated into species of animals. They are normally man-shaped with a head of the animal, and sometimes with a weapon. All of them have a wing-shaped protuberance, sticking out of their shoulder, probably symbolizing that they are angels, or holy messengers as they target survivors of the Akatsuki Incident, as they and other human possess a form of psychic that marked them as having the potential to become Agito. When they murder that certain type of person, they commit a ritual by using their hands, saying that they want permission to sin. At the same time, a halo appears over their head. Every one of them have their unique way of murdering, for example: leaving the corpse in a tree, dissolving the human, drying the human into death, changing the human into sand, pulling the human in the ground, dragging the human into the air and dropping them, vapourising, and so on. Killing a normal human is consider a taboo with a penalty of death.
OverLord of Darkness - OverLord (オーヴァーロード, Ōbā Rōdo?): He is the highest power among the Unknown, and is referred to as the creator of the human race. Long ago, there existed twin entities; one being the embodiment of light while the Overlord was embodiment of darkness. These twin 'brothers' co-existed in harmony for a time, eventually creating our world. However, neither could agree on who would reign over their creation, and a battle for control took place. In the end, 'Darkness' triumph over 'Light'. But with his dying breath, Light bestowed his essence upon humanity--the power of AgitΩ--with the hopes that, one day, they would evolve beyond his brother's control. But the Overlord refuse to let this occur, taking it upon himself to personally eliminate the vessels of AgitΩ. But even the murder of one of his creations provided too great a burden on him and thus created the Lords to act in his stead: Killing all humans who possess the potential to "evolve" into AgitΩ.
- Though he died around the Pleistocene, the Overlord left a puzzle that held a model of his very own DNA. Once the puzzle was solved, the Overlord's DNA was reconstructed from scratch. This allowed him to reform his physical body, starting off as a infant as he broke out of the containment box and escaped from the O-Parts lab. However, his birth was witnessed by Sakiko Mikumo, following her after he matured to a eight year old boy until she was killed by Anguis Femineus, whom the Overlord punished with death. Later on, as the Lords would go on to commit various supernatural murder of anyone reaching paranormal maturity, the Overlord underwent a rapid maturity to assume his adult form and watched from the sidelines as the AgitΩ Shoichi, along with Ryo and Makoto Hikawa, would complicate matters and interfer in his emissaries' work. Before long, casualties mounted on both sides, with paranormals and Lords alike suffering loses. But the lost of the El of the Sea opened the eyes of the Overlord to the reality that the vessels of AgitΩ, Shoichi Tsugami, Ryo Ashihara, and Kaoru Kino had grown too powerful. To this end, he would see to it personally that all AgitΩ were hunted down and stripped of the Seeds of AgitΩ. One by one, the Overlord claimed one Seed after another, until all three were rendered powerless--human.
- However, the essence of Light, radiating from the Seeds of AgitΩ, overwhelmed the Overlord as he vied for the Seeds' freedom. The Overlord would come to the revelation that only destroying the vessels, would the power of AgitΩ be vested; seeing no choice but to kill the riders. Shoichi, Ryo, and Kino would come to reclaim their power, and with Hikawa's aid, Shoichi dealt a decisive blow to the Overlord that released the Seeds of AgitΩ back to their rightful owners. The Riders would stand united and force the Overlord to retreat, for a time; but, the wound Shoichi dealt the Overlord was deeper than anyone would have thought. The shock that even normal human had grown strong enough to oppose his power convinced him that humanity was no longer worthy of existence, and so began orchestrating their extermination and start anew.
- In a month's time, the Overlord would put his genocide of humanity into action: relining the constellations of the Zodiac, ultimately killing off all of mankind in bizarre doppelgänger homicides starting with those born under Scorpio. To do this requires tremendous concentration, which would render him vulnerable. To this end, he creates the El of the Wind to be his bodyguard and the El of the Ground to continue killing those reaching paranormal maturity. But the Riders makes one final stand, and with the El Lords destroyed, the Overlord attempts to retreat but is defeated by AgitΩ's Shining Rider Kick. In the end, the Overlord survived and decides to postpone his plans and wait...to see what the future holds for humanity.

- El of the Water (水のエル, Mizu no Eru?, 33-35, 41-43): The strongest of the sea-based Lords, he used a forked spear as his weapon, along with telekinesis and regeneration. The Lord responsible for the Akatsuki Incident, "killing" Sawaki when he awakened as the 2nd AgitΩ. The El of Water allowed the others aboard to live until they undergo the evolution to AgitΩ, entering the body of Masumi Seika to ensure they never tell anyone of what had occurred. He eventually awakened, using Masumi to kill for him until he found the Overlord who gave him the power needed to leave his host, killing her in the process. Using Orcinus to stall AgitΩ, the El arrived to personally finish the job and kill Shoichi, who was intimidated by the El. However Ryo intervened and got Shoichi out of harm's way. The El of Water later encountered G3-X, yet spared him after sensing Hikawa's zero potential of becoming an AgitΩ himself. The El Lord was eventually defeated by AgitΩ (Burning Form)'s Burning Bomber, returning to the Overlord's body to recuperate and undergo a evolution of his own. By the time he completed the changes, the Overlord sent the El of Water after the AgitΩs. In his new Strenghthen Form, the El of the Water possess a halbred and heighten psychic power. He managed to overpower the Riders, taking out Agito while fighting the others and winning. But once Shoichi awaken with his memories restored, he battles the El in Burning Form after G3-X was injured. Soon enough, the tables turned and the El was wounded by Exceed Gills's Exceed Heel Claw and Another AgitΩ's Assault Kick before AgitΩ assumes Shining Form lands the deathblow with Shining Rider Kick. The El of Water was unable to return to the Overlord's body in time and was reduced to a puddle of water to his creator's shock.
- El of the Wind (風のエル, Kaze no Eru?, 47-51): The strongest of the air-based Lords. Created by the Overlord to ensure no one intervene while he is channeling his power, The El of the Wind uses a bow to fire arrows at his victims, causing them to disappear with only their clothes remaining. The El of the Wind fought Gills after killing off a witness to the Overlord's trance, overpowering him in even Exceed Form and AgitΩ's Burning Form with little trouble. He later aided the El of Ground in defeating AgitΩ Shining Form. But he fought Exceed Gills and G3-X, wounded by the GX Launcher and killed by Exceed Gills's Exceed Heel Claw.
- El of the Ground (地のエル, Chi no Eru?, 49-51): The strongest of the land-based Lords, he was created by the Overlord to continue the Lords' task to slaughter those with the potential to become AgitΩ. A sword-wielding El, he was able to turn people into dust with his sand and borrow underground. This El holds considerable strength as apparent from his first encounter with Gills, who engaged him in battle for the death of Risa. He was even able to outmatch AgitΩ, despite switching into his Ground and Burning Forms. However, he was no match for Shining Form AgitΩ, whom mortally wounded him with his Shining Clash attack. The El was then renergized and Strengthened by the Overlord, but he was still outmatched by Shining AgitΩ and destroyed by AgitΩ's Shining Rider Kick.

Once partially awakened, the El orders him to kill Majima and Mana. But Orcinus is intercepted by Agito, but managed to overpower him. However, Orcinus was halted by Gills arriving after his revival. The Lord attempted to retreat, but had to fight both Gills and G3-X until he managed to retreat. Orcinus then attempted to kill Shoiuchi, forcing him to fight the Lord as Agito with Ryo present. The fight ended with Agito Ground Form's Rider Kick destroying the monster.
- Propheta Cruentus (プロフェタ・クルエントゥス, Purofeta Kuruentusu?, 34): A Mantis Lord (マンティスロード, Mantisu Rōdo?), he uses twin swords as his weapons. Cruentus attempted to kill Majima, but Gills interverned and battled the monster until the El of Water arrived to exterminate Gills. The Mantis Lord later attempts to kill Majima, only for G3-X to intervene this time. Cruentus soon goes to attack Gills directly, with G3-X interfering until the El of Water evened the odds. However, Shoichi arrived with a renewed sense of courage, assuming Burning Form as a result. With his newfound powers, Agito obliterated Cruentus with Burning Rider Punch.
- Scarabaeus Fortis (スカラベウス・フォルティス, Sukarabeusu Forutisu?, TVSP): A Beetle Lord (スカラベロード, Sukarabe Rōdo?) who uses a mace and shield, his method of killing to shoot a needle from his horn at his victim in the neck, turning them to ash. His first victim was Susumu Murano, targeting the younger brother and the bodyguard assigned to him. He was intercepted by G3-X, overpowering both him and Agito before retreating. He later resurfaces and battles Agito Burning Form, forced to retreat. When Beetle resurfaces, he defeats G3-Mild and overpowers G3-X until Omura regains consciousness installs his suit's battery to fully reenergize G3-X. Gills and Agito join the fight then, with Agito assuming Burning Form to battle the Lord. The three Riders were no match for the Lord until Agito assumes Shining Form for the first time, killing the Lord with his Shining Clash attack.
- Volucris Ulucus (ウォルクリス・ウルクス, Worukurisu Urukusu?, 43-45): A Owl Lord (オウルロード, Ōru Rōdo?), the method of killing is cracking the skulls of their victims. Occurring between episodes 40 and 41, Shoichi encounters find a teenage girl name Sayaka who have a supernatural ability and can control it by bracelet, but unfortunenately she drop the bracelet and the wear by Mana who also have Supernatural ability, and it makes Risako Fukami of the military group GA interest on her power for enhacing the power of G4 by using the supernatural ability to perfect the designs she stole from Ozawa. Soon, while Agito and Gills deal with the AntLords that attack even normal humans, G3-X enters a final showdown with G4.
