Nocodazole is an anti-neoplastic agent which exerts its effect by depolymerizing microtubules. Microtubule formation is an important structural feature of cells as they enter mitosis. Several drugs including vincristine and colcemid, are similar to nocodazole in that they interfere with microtubules and cause arrest in G2/M phase of the cell cycle. As nocodazole affects the cellular cytoskeleton, it is often used in cell biology experiments as a control: for example, some dominant negative Rho small GTPases cause a similar effect as nocodazole, and constitutively activated mutants often reverse or negate the effect. Also, nocodazole is frequently used in cell biology laboratories to synchronize the cell division cycle. Cells treated with nocodazole enter mitosis but cannot form metaphase spindles because microtubules (of which the spindles are made) cannot polymerise. This activates the spindle assembly checkpoint, and the cells arrest in prometaphase. For cell synchronization experiments, nocodazole is usually used at a concentration of 100ng/mL of culture medium for a duration of 12-18 hrs. ... My research has centered on the experimental investigation of regulatory mechanisms in ovarian function, especially on the control and cellular mechanism of oocyte meiosis using amphibian, insect and teleost ovaries as experimental model systems.
The research has focused on the effect of steroids and cytoskeleton-active agents such as taxol, nocodazole, soluble acrylamide and colcemid on the processes of nuclear migration within the oocyte and dissolution of the nuclear envelope during meiotic cell division (prophase I - metaphase I transition).
This work employed molecular approaches including the use of monoclonal antibody probes for cytoskeletal proteins, Western blotting, confocal immunofluorescence microscopy, and morphological analysis at the ultrastructural level using immunogold procedures with transmission EM.
