When his father was elevated to the papacy, as Paul III, in 1534, Pier Luigi was named Captain-General of the Church. His father later made him Duke of Parma and Piacenza, properties that had previously been a part of the Holy See. He ruled these cities tyranically, and the aristocracy of Piacenza murdered him in 1547.
Nel 1368 Nicolò Farnese, dopo l'attacco del prefetto Giovanni di Vico, portò in salvo il Pontefice Urbano V prima nella Rocca di Viterbo e, successivamente, in quella di Montefiascone.
PierLuigi fu sgozzato ed il suo corpo venne gettato nel fossato sottostante il palazzo.
Alessandro Farnese, primogenito di PierLuigi, nacque nella Rocca di Valentano il 7 ottobre 1520.
