Ribonucleoprotein(RNP) is a compound that combined ribonucleic acid (RNA) and protein together. It is one of the main components of nucleolus. RNP is an RNA binding motif in an RNA binding protein. Aromatic amino acid residues in this RNP motif result in stacking interactions with RNA. Lysine residues in the helical portion of RNA binding proteins help to stablize interations with nucleic acids. This nucleic acid binding is a result of highly positive nature of lysine side chains and the negative character of nucleic acid phosphate backbones.
Unlike cytoplasmic ribonucleoprotein particles which are potassium-dependent (2), these nuclear particles require a sodium environment for their metabolic activity in the nucleus; furthermore, unlike cytoplasmic particles (3,4), the intranuclear metabolism of these nuclear is resistant to preincubation of the nuclei with ribonuclease (RNAase) but sensitive to pre-incubation of the nuclei with deoxyribonuclease (DNAase).
Because of the known intimacy of the metabolism of protein and RNA in protein synthetic processes in the cytoplasm (3, 22), it was of interest to study the simultaneous intranuclear metabolism of protein and of RNA in the nuclear ribonucleoprotein particles of one individual.
This is in marked contrast to the behavior of cytoplasmic ribonucleoprotein particles, whose protein metabolism is increased by potassium and decreased by sodium (2).
In this study, we used bisulfite sequencing to determine the methylation dynamics of the imprinted genes small nuclear ribonucleoprotein N (Snrpn), insulin-like growth factor 2 receptor (Igf2r), mesoderm-specific transcript (Mest; formerly Peg1), paternally expressed gene 3 (Peg3), and H19 fetal liver mRNA (H19).
Paternal specific expression of the known imprinted genes SNRPN (small nuclear ribonucleoprotein- associated polypeptide N gene) and IPW (imprinted gene in the Prader-Willi syndrome region) was maintained in the A9 hybrids.
We also studied the allelic expression of the small nuclear ribonucleoprotein polypeptide N gene (SNRPN), which is reportedly maternally imprinted in humans, and compared the imprinting status with that of IGF2.
