Route 249 is a short, mostly two-lane highway running from Business US 71 in Carterville and ending at Interstate 44 in Joplin. The route currently detours from its intended path at Zora Ave, between Joplin and Carterville. Construction is set to resume shortly on a full limited access highway to connect Business 71 (Joplin) to I-44 as a bypass of Joplin. The road is being built into a freeway and is planned to be a future part of Interstate 49 which has been proposed to run down the U.S. Highway 71 corridor. Route of the highway

The highway begins at an interchange near the city hall of Carterville in a concurrency with Missouri Supplemental Route HH. At about a mile, it turns south in a concurrency with Supplemental Route AA (Prosperity Road), and two miles further, turns west on Supplemental Route VV (Zora Road) very briefly. All of these supplemental routes were in existence prior to Missouri 249 and Missouri 249 was routed with them as it was extended north. The road runs on a straight course south and has an interchange with Route 66/Business I-44 before reaching the interchange of Interstate 44/US 71 where the road ends.
